SPM and AFNI are quite different in many ways. Although you can do linear regression modeling with both of them. However the interface and implementation of the methods can differ significantly. It is impossible to answer your question concisely (and in an unbiased manner). Better to go through the websites for SPM and AFNI and look at their demos and descriptions so that you can make up your mind about what to use.
