If I understand you accurately, you have a model like the following

Connectivity with seed B = a0 + b1 * A + b2 * C + b3 * A * C + b4 * movement + b5 * age + b6 * gender + e

Yes 3dttest++ can take care of the analysis. If you are not interested in the parameter a0 in the model. You don't need to worry about centering.

No, you don't need to run separate analyses. One analysis should give everything you want. The main effects of A and C are just b1 and b2 in the model.
