You don't really need to since you can specify whatever hemodynamic response function you'd like to put in the model.

I'm not so sure what kind of stimulus timing you have, but for the purpose of creating the connectivity regressors one option is that you can manually create those files of 1/0/-1 (and you don't need to worry about local versus global time).
